Test organism은 histidine-negative(his-)인 Salmonella typhimurium의 영양요구체(auxotroph) 균주이다. 그래서 Ames test에서는 bacterial system이 mammalian test system에 더 근접하도록 하기 위해 activating enzyme의 source로서 liver homogenate S-9를 넣어준다. Chemical의 mutagenic effect는 종종 organism의 enzymatic pathway에 의해 influence(영향)을 받아 nonmutagen이 mutagen으로 전환되거나 혹은 반대로 mutagen이 nonmutagen으로 전환되기도 하는데, 포유류에서는 이러한 toxification이나 detoxification이 간(liver)에서 일어난다.
